An important part of the sampling is the determination of the place of sampling in order to maximise the chance of capturing DNA from the target organism. In doing so, the ecological wishes of the organism to be determined must be taken into account. The chance of detection of a species can be greatly increased by taking multiple samples in different places in one water body. DNA is quickly broken down (within 2 days at a water temperature of 20 degrees Celsius). The whole procedure and tips are explained in the protocol beloning to environmental sampling set.

Environmental sampling set

Sylphium molecular ecology has compiled a special sample set for sampling water bodies for eDNA determinations. Water samples are filtered on site over a filter unit. This method has the advantage that the chance of contamination between samples is minimized and that only a few materials has to be taken into the field for sampling and filtration. In addition to a filter unit, the sample set also contains a disposable 60 ml syringe. With this syringe, the sampler can take a water sample, without touching the water. This will prevent cross contamination between water bodies. The filter unit is then placed on to the syringe and the water is pressed over the filter unit. A vacuum pump is not necessary for this. After adding the preservative liquid and closing the filter unit with supplied caps, the water sample remains stable for a long time (a few months) without cooling. The filter unit can then be sent to the lab of Sylphium molecular ecology for further analysis. Sample sets can be ordered in the webshop. Sampling rod

Sylphium molecular ecology has developed a special sample rod for sampling hard-to-reach places, for example overgrown and deep banks. The 60 ml sterile disposable syringe (from the sample set) is placed in this rod, after which the water is drawn up. By using this rod, no waders, worth suits, a boat or other tools are needed to take water samples. This prevents DNA contamination between different sampling locations and significantly speeds up the sampling time. After sampling, the syringe can be replaced and the rod is ready to be used again. Sending samples

The filtered water samples in the filter units can sent unrefrigerated to the lab of Sylphium molecular ecology. For this use, a bag is supplied with a unique sample number. Based on this number, the sample can be identified afterwards. The samples can be sent (registered) via regular mail to:
